Urban legend has it that Curt can do it with the tank on, but I havent seen it yet on Myth Busters

He props the seat end of the tank up so that he can remove the needle assembly!!!!!
I just remove the tank, it doesnt take that much longer!!!!!

I am hoping one can delete ones question before others see it I told her you have to remove the seat.Believe it or not I actually ride on two wheels upright!

You don't have to remove the seat or the tank to remove the main jet. I have done it several times.

It is easier for me to jack the bike up with a bike jack, put some kind of light on the opposite side of the carb between the cylinders so you can see the carb bowl screws and take them out. You can then get to the main jet with no problem.
