Without a repeat performance, and then a complete fill up, it'll be tough to tell.
I hate these kind of things.

Just curious, are you running an inline filter?

Yes, a Pingle.

One question... is the Fuel line going below the manifold?

If so, that may be your problem.

I run the HSR42, stock petcock, a Yost power tube, a filter provided by Shane at SS Customs Cycles with the HSR42 kit and I have my fuel line *OVER* my manifold. Last year, the furthest I got with a trailer about 155 miles when my light came on... about another 3 miles I switched to reserve. I ended up at about 4.2 gallons total fill up at 165 miles.

I didn't drill out the vent tube, nor remove my rollover valve and my gas cap is not vented.

I think ~38 miles per gallon (GPS miles not odometer miles) is pretty dang good at 65MPH with a trailer.

I think my Yost power tube jet access extension might have something to do will it... making the bowl deeper. I don't know for sure about that, since it is just speculation.

Mine comes on, then I switch to reserve. Usually have around 35 miles or so before she's empty, but I find a gas station just as soon as I can.

Nope, running filter and line above manifold also. Also running Yost Power Tube. On my last tank, I averaged 42 mpg (with fairing running 70-75 mph on the superslab).

I'm going to run this tank down and force a repeat performance if possible.

Your new petcock pickup should be the same height as the stock petcock
you remove, otherwise the pickup level changes.
