Yes, it could cause the bike to die, and then start again easily, as it is replenished when the engine is off. Here is another easier way to check the level..... http://weislake.com/sig/mucker/carb/carbfloat/carbfloat.htm You should be using the 04 and later spec, not the 99-03. Yamaha made that change.
